Assay Conditions: 50 µL reaction mix (50 mM HEPES pH 7.4, 50 mM NaCl, 200 µM CaCl2, 0.05% Tween-20, 0.1% BSA, and 0–1500 ng biotin-labeled PCSK9) is added to wells precoated with LDLR Ectodomain. Incubate for 2 hr. Add streptavidin-labeled HRP and incubate 1 hour. Finally, add HRP chemiluminescent substrates and read luminescence.
Formulation: 40 mM Tris, pH 8.0, 110 mM NaCl, 2.2 mM KCl, and 20% glycerol
